1941, Slap Shot, and Animal House are tough to beat in terms of straight up comedy. But my favorite is probably a little-known 1980's dramedy called Tin Men with Danny DeVito.
I really enjoy any movie produced by Mel Brooks. One of my favorites is Young Frankenstein, but Blazing Saddles is in the running. I also enjoy a variety of movies that Billy Crystal is in.

True Lies. With Jamie Lee Curtis a lot of comedy movies border on stupid and I just don't find them that funny. But True Lies gets me laughing every time.
