semen will fluoresce under black light. so will cat piss (only way to find actual target areas on the carpet) and many other substances. the thing is that most things fluoresce at a different level of light, and i think semen shows up brighter that, for example, cat piss. plus your cat probably doesnt piss on your crotch. plus you have spatter patterns, that can tell you more. sounds like semen to me though.
i think to test it they use a swab dipped in a liquid, it may be a neutral medium (just to soften enough to get the sample) or a reactive chemical (turns a pretty color if its the reactive substance) im not sure of actual procedures, of course.

definately try to keep the stain intact, try not to fold through it because it could fall apart. sealed bag like was already suggested. you know were here for ya check.
