No. If you maintain stimulation long enough you will ejaculate in reverse, into your bladder. Medical professionals call this "a retrograde ejaculation". No harm will be done.
A penis's knob will possibly get irritated after a certain amount of continuous stimulation & get very sore to touch.
As others have said I don't think you'll cause any real harm. However that said I'll tell you this: Masturbation to the point of almost coming depending on how long and "how" you do it can lead to two things... One is a massive money shot and the other is a buildup to the point of cloggage. Neither are dangerous from what I know but the latter while it may be more enjoyable during the mid-cycle can lead to some discomfort as you pretty much clog up the pipes and if / when you do come there tends to be just a dribble. Honestly though I think it's more about "how" you do it. If you get near the point of cuming and back off immediately you're okay. If you build up to that point where the wave breaks and then feverishly try to not cum and do that repeatedly or ride that cusp teetering on cuming or not then that tends to clog things up. You won't "clog up" anything. "Edging" to the point that you no longer feel the orgasm is a "retrograde" orgasm where you ejaculate backwards into your bladder. If you edge to the point of a reverse (retrograde) orgasm and then continue to masturbate because you want to feel it, the sensation is weak because you already ejaculated... in reverse. No harm at all will be done in any case. If you edge for a while but not to the point of either an orgasm or a reverse orgasm (no orgasm at all) then the semen will be re-absorbed and will not harm anything that way either.
Actually it can cause harm, it's rare, but possible that things backed up repeatedly can lead it irritation and thus infection if you suppress an ejaculation. Can lead to seminal vesicle infections, called Epididymitis, because that area of the body is irritated and a pathogen might have an easier time getting in there and causing an infection.

You can't masturbate 'too much' - your body simply won't let you. When it needs a rest your penis will remain flaccid & you won't be able to do a thing with it, apart from pee. Seminal fluids which, as previously stated, mainly consist of water, don't take long to accumulate, and as sperm take longer to generate than semen does, the sperm count in each consecutive ejaculation made within a short period will obviously have a lower sperm count & will tend to become more & more watery.
