Hey hey what’s going on everyone!? It’s closing in on 15 days of incubation and my casings are just about completely covered. My question is when it fruits, what would be the best way to get new spores to continue my growing? Patrickbull gave me great advice on how to make spore prints but is there any other way then making a spore print? Let me know what’s up if there are.

Wanna take a spore print?? Here's one of many great methods! (this is what i do)

Go to the store and buy those tiny Glad-ware Mini Rounds. They come in packs of 8 for like 2$. Perfect size for spore prints.

Each cup is 1/2 cup volume.

Fill a brand new empty cup half full of water, leave the lid open just a crack (so it doesnt explode), and put it in the microwave for 2 mins.... the water will start to boil and create steam, sterilizing the cup and lid.

Quickly take the cup out of the microwave and dump out the water (a little bit will remain but its no big deal). Snap the lid on tight and wait for the cup to cool (with the lid on). Put this in your clean glovebox.

Get a fresh young mushroom cap and transfer the cap to glovebox or another sterile environment. If you dont have a glove box, you can attempt a super fast transfer inside of your fruiting chamber, since its probably a little cleaner than the room your growing in. (your choice. but id recommend a glovebox. I have a fan on the side of my glovebox which sucks filtered air from the outside to the inside, so when im working with my glovebox i always let the fan run for about 5 mins before i start transfering or inoculating anything. Theoretically, the filtered air should displace the air that is initially inside the glovebox, forcing it through the tight but imperfect seal of the rim on my glovebox lid.)

If you do have a glovebox, you will have to find a clever way of transfering the cap to your glovebox with MINIMUM exposure to air or other sources of contam. Since i dont know what everyone has, i cant tell you what to do here.

IMPORTANT: DONT even touch that cap with your hands prior to printing. I have lab tools that i flame, one for stabbing the cap (to hold it steady) and a scalpel for cutting the cap loose from the stem. Once the cap is loose from the stem, transfer it asap.

When everything is ready, open the lid of the sterile Glad-cup, and put the cap in (gills down). Then, seal up the lid. The print will eventually appear on the bottom of the cup. The next day, take the cap out (in the glovebox) and then re-seal the lid. I tape it shut just to make sure.

Seriously... this works like a charm.

I didnt go through all the steps for cleanliness and sterility, but it should be obvious... wink.gif

I use this technique for all my prints, and every time ive made syringes out of me prints it had worked perfectly.

Not only that, but you can label and keep track of all yr prints quite easily. Ive also (recently) revived prints that were over a year old... total success.

So you've made a print! Its the best way to store those genetics for a longer period of time!

BUT now you want to use those genetics.
I find that making a spore syringe is effective, and is really easy if you have followed the above directions for making a spore print using glad-ware cups. Here's how:

Get another Glad-cup and fill it 2/3rds full of water (distilled preferably). Put it in the microwave and boil for 2 mins (pretty much the same as before). Except, this time you will want to shut off your microwave and leave the door closed. Sit there and watch until the water stops boiling, then wait a 1 min. Remember, even though the lid is open, the inside of the microwave is very clean (not quite sterile though... some thermophilic bacteria will not be dead.) Microwaves have been used to sterilize things in the lab before (granted, its not quite the same as your home edition, but it still works well enough that i havent had problems).

Open the door and shut the lid of the cup quickly.
Put this hot cup of water in your glovebox, along with....
1. one (or more) brand new sterilie syringes w/ needles. I use 10ml plungers with 13 gauge needles.
2. An alcohol burning lamp (and lighter)
3. The spore print of your choice

Each spore print will make upwards of 5 (or more) 10ml syringes. I usually only make 2 syringes, because im not that concerned with having massive amounts of spore syringes around. I wouldn't use them all anyway!

Take a new syringe/needle combo out of its wrapper and assemble it together. Flame the tip of the needle and proceed to stab it directly through the lid of the cup containing the sterilized water you just made. Suck up 10cc's of water into the syringe. Next, take the needle out and flame it again. Proceed to stab through the lid of the cup containing the spores. Shoot out all of the water into the cup. NOTE: you must wait until your hot water has cooled to room temp or you will risk destroying your spores!

When the water is in the cup, it is re-hydrating the spores. I like to wait for 15 mins before i suck the water (that now contains many spores) back up into the syringe.

Pull out the needle and flame it again. Then put the protective cap over the needle and secure it tightly.

You have just made a spore syringe!!

Like i said before, you can make a lot of syringes with one spore print. So, if you may need to adjust your technique slightly to accommodate the number of syringes you are making. (think about how much sterile water you need for 5 syringes... )

Instead of putting the spores into the dry glad cup, then transferring, suspend the cap with a homemade stand made of paperclips over top of the sterile water. Skip a step and a possible contamination.
